Type of Human Subjects Involvement:
The objective of this project is to construct normalized cDNA libraries from human tissues as a resource to the community to expedite gene identification and mapping. The identities of the human subjects are not known by the PI, thus assuring confidentiality. Total cellular RNA extracted from human tissues (obtained for purposes other than those of this project) is used for construction of cDNA libraries that are directionally cloned in a phagemid vector. cDNA libraries are then normalized according to procedures that we have developed (Soares et al., 1994: PNAS 91,9228-9232; Bonaldo, Lennon and Soares 1996: Genome Research, in press). In addition we are taking advantage of subtractive hybridization strategies to reduce the representation of already identified (arrayed and sequenced) I.M.A.G.E. clones from libraries yet to be surveyed. Moreover, we are attempting to develop methods for construction of full-length cDNA libraries.
